This project was a scientific substudy associated with the NCI's longterm followup of women exposed in utero to the high-dose estrogen diethylstilbestrol (DES), and matched controls.  Dr. Palmer is the PI of all studies concerned with breast tissue. Our goal was to investigate whether prenatal exposure to DES leads to characteristic genetic abnormalities in breast cancers and premalignant lesions.

“Genetic Abnormalities Early on the Path of Breast Tumorigenesis"

Histologically normal human breast epithelium from three groups (reduction

mammoplasties, sporadic breast cancer surgeries and BRCA1 or 2

constitutionally heterozygous women) will be examined in order to compare

the rate and sites of LOH. Candidate tumor suppressor genes, located at

frequent sites of LOH, will be identified and their relevance to early

stages of breast tumorigenesis tested using immunohistochemistry and in situ

hybridization.
